Acquired Immunodeficiency Syndrome (AIDS):

• Attaches to CD4 receptors on host cells and enters them.

• Uses CCR5 chemokine receptors early in infection; macrophage tropic.

• Uses CXCR4 chemokine receptors late in the infection; T cell tropic.

Mechanism:

Nef gene downregulates MHC I.

Tat gene inhibits cytokine synthesis.

• Multiplication in lymphocytes and macrophages.

• Cytopathic effects on lymphocytes and macrophages.

• TH destroyed.

• Antigenic drift and glycosylation of gp120.

• Immune deviation toward TH2 response.

Macrophages and Dendritic Cells:

• Infected with HIV but resistant to destruction.

• Spread virus to lymphoid organs.

• Have impaired AP function.

Antibodies generated against HIV; used for diagnosis in ELISA, western blot, etc.
