What is the mechanism of action of tumor-killing cells?

Tumor-killing cells are of two types: cytotoxic T lymphocytes (CTLs) and natural killer cells. If the MHC class I antigens covering the CTLs were lost, the CTLs would lose their anti-tumor activity. Then, the NK cells would be stimulated alone. Tumor antigens are presented in the groove of class I MHC molecules and are recognized by CD8+ T cells. Thus, loss of class I MHC would prevent the tumor cell from being destroyed.
